Our lab explores various aspects to make Hyper_CEST MRI even more efficient. One strategy are Xe host structures that can bind many atoms unlike the first generation hosts such as cryptophane-A.
However, these individual hosts partition easily into phospholipd bilayers due to their hydrophhobic nature. CEST spectroscopy shows enhanced saturation transfer in the lipid environment. This can be used to prepare liposomal and micellular nanocarriers for selective cell labelling (brain endothelial cells of the blood brain barrier in this case) with excellent conditions for Hyper-CEST MRI.
In an alternative appriach, we explore the gas exchange properties of biogenic structures such as bacterial gas vesicles. These show both accelerated exchange and a capacity for thousands of Xe atoms per nanocarrier to yield unprecedented efficiency in CEST signal build-up.
